General Description

provided by Cybertruffle
Colonies. Growth rate is 4.45 cm diameter, aerial mycelium floccose, white to pale red and pigmentation dark red on Potato Sucrose Agar (PSA) plates after 4 days. On Potato Dextrose Agar (PDA) slants after 10 days: Growth rate 7.75 cm length, with floccose, white to pastel red aerial mycelia and reddish brown pigmentation. "Chlamydospores" sparse, produced singly, in chains or in clusters. The one isolate tested of F. acuminatum was able to grow on Glycerol Nitrate Agar (G25N) medium with a growth rate more than 2 cm diameter and produce white aerial mycelium and greyish yellow pigmentation. On mannitol medium growth rate is less than 4 cm diameter, producing pale yellow mycelium and reddish yellow pigmentation. On Czapek-Dox Iprodione Dichloran Agar (CZID) medium growth rate is more than 5 cm diameter, producing pale orange aerial mycelium and greyish yellow pigmentation. On tannin-sucrose agar (TSA), producing urease and phosphatase enzymes. Peroxidase and pyrocatechol oxidase enzymes, acid on creatine-sucrose agar (CSA) and acetylmethylcarbinol compound not produced. Conidia. "Macroconidia", strongly curved into a sickle-shape, 3- to 7-septate, but commonly 5-septate, 30-60 × 3.5-5.5 μm, with elongate apical cell and distinct foot-shaped basal cell. "Microconidia" sparse, fusiform, 0- to 1-septate, 10-15 × 2-4 μm, produced singly or in pairs from conidiogenous cells with only one active fertile point. Ascomata. Perithecial, known only from the mating experiments by W.L. Gordon, resulting from inoculation of opposite mating strains on to sterile wheat straw standing in water in flasks, mature in six to eight weeks at 22°C, scattered but tending to form around the nodes from a weak byssus, deep purple, appearing black, obpyriform to ovate with a smooth outer wall and undergoing a pinched lateral or pomiform collapse on drying, 190-210 μm diam.; in section the outer wall 25-33 μm thick, consisting of 6-7 layers of oblong to globose cells 7-12 × 4-6 μm; the inner wall consisting of 2-3 layers of very thin-walled, compressed, colourless cells; locule filled with a delicate pseudoparenchymatous tissue in early stages of ascus development, but this soon disintegrating as asci enlarge. Asci. Clavate, shortly stipitate, with a thin undifferentiated wall, 4- to 8-spored. Ascospores. Smooth, colourless, ellipsoidal to oval but becoming slightly narrower at the central septum, 13-20 × 6-7 μm, often remaining 1-septate although occasionally a second septum developing in the somewhat larger upper cell.
