Euglena gracilis ("schlankes" Augentierchen) ist eine Art der Protisten aus der Gattung der Augentierchen (Euglena).
Euglena gracilis ist 35 bis 65 Mikrometer lang und 5 bis 15 Mikrometer breit. Das hintere Ende der schnell schwimmenden Zellen beschreibt einen weiten Kreis bei der Eigendrehung, die besonders stark wird, wenn die Zelle nicht mehr schwimmt.[1]
Die sechs bis zwölf abgeflachten, schildförmigen Chloroplasten sind groß und weisen zu beiden Seiten des mittigen Pyrenoids uhrglasförmige Paramylon-Hauben auf. Palmellae entstehen im Schlamm.[1]
Euglena gracilis ist eine seltene Süßwasserart, die in Gräben und Teichen mit verrottenden Blättern vorkommt.[1]
Euglena gracilis ist ein bedeutender Modellorganismus insbesondere in der Chloroplasten-Physiologie, da Kulturen jahrelang ohne Licht gelagert werden können, aber rund 24 Stunden, nachdem sie dem Licht ausgesetzt wurden, wieder mit der Photosynthese beginnen. Einige Stämme (es existieren auch chloroplastenlose) finden Verwendung bei der Erforschung unter anderem von DNA und RNA, heterotropher Ernährung, Photorezeption und zum Fettstoffwechsel.[1]
Euglena gracilis-Exemplare sind außerdem Teil der Satellitenmission Eu:CROPIS des DLR. Zum Test eines bio-regenerativen Lebenserhaltungssystems wird in Miniaturgewächshäusern im Satelliten künstlicher Urin durch Bakterien in Nitrat umgewandelt. Die Euglena bauen das dabei entstehende Ammoniak ab. Mit dem Nährstoff werden Tomatenpflanzen aufgezogen. Der Test findet bei durch Rotation erzeugter künstlicher Schwerkraft durchgeführt, nacheinander bei Mond- und Marsbedingungen.[2]
Euglena gracilis dient außerdem als Testorganismus für Vitamin B12, da es dieses nicht selbst synthetisieren kann.[3]
Euglena gracilis lässt sich im Gegenteil zu den meisten anderen Euglena-Arten und Augentierchen sehr einfach kultivieren. Dafür wird ein Ansatz von Euglena in ein Erdabkochungsmedium mit einem Stückchen Hartkäse versetzt. Damit eine solche Kultur gelingt, muss die Erde allerdings alle nötigen Nährstoffe enthalten und sollte außerdem nicht mit Pestiziden versetzt oder kürzlich gedüngt worden sein.[3]
Für den Einsatz in Lebensmitteln und Nahrungsergänzungsmitteln in der EU ist Euglena gracilis in Form von Pulver zugelassen.[4] Sie wir als Quelle für Beta-Glucane genutzt.[5]

A morphological and molecular study of the Euglenozoa put E. gracilis in close kinship with the species Khawkinea quartana, with Peranema trichophorum basal to both,[4] although a later molecular analysis showed that E. gracilis was, in fact, more closely related to Astasia longa than to certain other species recognized as Euglena. The transcriptome of E. gracilis was recently sequenced, providing information about all of the genes that the organism is actively using. They found that E. gracilis has a whole host of new, unclassified genes which can make complex carbohydrates and natural products.[5][6]

Mixotrophe facultatif avec dominance du mode hétérotrophe, il peut se multiplier en pure autotrophie ou à l’obscurité en hétérotrophie en présence de nutriments organiques. Dans ce dernier cas, on observe une régression des chloroplastes (micro-organismes présents dans les cellules eucaryotes photosynthétiques (plantes, algues) et sensibles à la lumière) disparition des pigments photosynthétiques et décoloration de la cellule.
À la lumière, la présence des chlorophylles a et b, caractéristiques des lignées algales « vertes », indique que cette espèce a acquis la capacité d’autotrophie par une endosymbiose (coopération mutuellement bénéfique entre deux organismes vivants) secondaire. La multiplication s’effectue par fission binaire (= scission longitudinale).
La culture in vitro confirme la grande souplesse d’adaptation d’E. gracilis : croissance à des pH inférieurs à 2, temps de génération quasi identique pour des pH entre 4 et 9 (moins de 11 heures en mode mixotrophe), faculté d’utiliser de nombreux substrats organiques comme source de carbone et/ou d’azote, grande résistance à des concentrations élevées en métaux lourds. E. gracilis peut se révéler un bon candidat pour la production de biocarburants, moins performant par exemple que Botryococcus braunii pour ses teneurs en lipides, mais bien plus rapide pour ses temps de doublement.
Le 20 décembre 2018, l’entreprise Kemin Foods L.C. fait la demande qu'Euglena gracilis soit reconnu comme Novel Food. C’est le 25 mars 2020 que L’EFSA (European Food Safety Authority) autorise la mise sur le marché européen d'Euglena gracilis en tant que Novel food. Le groupe scientifique considère que les cellules entières séchées d'Euglena gracilis, sont sans danger pour les utilisations proposées. Euglena gracilis est proposé en tant que complément alimentaire pour le contrôle du poids et en tant qu'ingrédient alimentaire ajouté à des produits tel que des barres protéinées, des produits laitiers, des jus de fruits ou des boissons substituts de repas.[2]

La membrana cellulare di natura glicoproteica è molto flessibile, visto che la cellula cambia forma in base anche agli stimoli ricevuti. Infatti il corpo può assumere una forma sottile e lanceolata dalla lunghezza massima di 100 µm, ma anche quella di una sfera dal diametro di 20 µm. L'Euglena gracilis è dotata di due flagelli, ma in genere solo uno di questi fuoriesce dal reservoir e può pertanto essere definito emergente. Il reservoir (o tasca), che ospita i due flagelli, è localizzato sull'estremità anteriore. Il flagello emergente ha funzione locomotoria e presenta mastigonemi.[1]
Nella tasca c'è una macula fotorecettrice, al di sopra della quale si trova una struttura ricca di pigmenti (stigma), che forse è anch'essa implicata nel processo di percezione della luce. Grazie a questa macula la cellula è in grado di muoversi verso la luce per favorire il processo di fotosintesi clorofilliana. Secondo alcune ipotesi, la massa sarebbe distribuita asimmetricamente nella cellula (il nucleo sta generalmente nella parte inferiore) e ciò ne determinerebbe l'orientamento all'interno dell'acqua. Successivamente si è invece supposta l'esistenza di un gravirecettore che agirebbe mediante canali ionici sull'allineamento, che tra l'altro muta a seconda dell'età dell'organismo.[1]
Essendo dotato di cloroplasto, è in grado di svolgere la fotosintesi clorofilliana. Tuttavia, se viene posto al buio ed a contatto con fonti di carbonio organico, è in grado di digerire i propri nutrienti, anche grazie ad un vacuolo contrattile che conserva enzimi oltre ad avere una funzione osmoregolatrice. Non solo il differenziamento dei proplastidi in cloroplasti dipende dalla presenza di luce, ma la cellula è in grado di ritrasformare questi ultimi in proplastidi per comportarsi come un organismo eterotrofo.[1]

Euglena gracilis jest przedstawicielem kladu Euglenid, który razem z Kinetoplastida, Diplonema i Symbiontida tworzą grupę Euglenozoa, należącą do supergrupy Excavata.
Euglena gracilis należy do rodziny Euglenaceae, która jest monofiletyczną grupą jednokomórkowych wiciowców, prowadzących wolny tryb życia, żyjących głównie w środowiskach wodnych. Ich cechą charakterystyczną jest znajdująca się pod błoną komórkową pellikula, stanowiąca powłokę zbudowaną z pasków białek. W przypadku helikalnego ułożenia pasków, uczestniczą one w specyficznej dla Euglenacae formie ruchu, polegającej na zmianie kształtu ciała (metabolia).
Rodzaj euglena skupia jednokomórkowe organizmy o niespłaszczonej budowie, charakteryzujące się zdolnością do dynamicznych ruchów i dużymi chloroplastami. W obrębie rodzaju euglena wyróżnia się podgrupy na podstawie kryteriów morfologicznych. Euglena gracilis należy do grupy euglen o brzeżnie ułożonych chloroplastach z pojedynczym pirenoidem. Do grupy tej zalicza się także m.in. Euglena hiemalis i Euglena longa[3].
Mitochondria E. graciliis pełnią swoje funkcje w różny sposób, w zależności od warunków. W środowisku tlenowym funkcjonują podobnie jak mitochondria w komórkach ssaków. W warunkach anaerobowych przeprowadzają syntezę kwasów tłuszczowych, prowadzącą do estrów wyższych kwasów tłuszczowych (wosków), które odbierają elektrony powstające przy glikolitycznej syntezie ATP i utlenianiu pirogronianu. Niektóre elementy szlaku beztlenowego w mitochondriach E.gracilis są również obecne w anaerobicznych mitochondriach niektórych bezkręgowców lub w hydrogenosomach występujących u niektórych beztlenowych protistów[4].
Do wzrostu Euglena gracilis wymaga witamin B1 i B12, które są pobierane aktywnie z użyciem energii ze środowiska. Poza tymi witaminami jest zdolna do syntezy innych witamin niezbędnych dla człowieka, takich jak C i E. Biosynteza witaminy C odbywa się na drodze specyficznego szlaku metabolicznego, łączącego cechy biosyntezy witamin u zwierząt i roślin[5].
Genom Eugleny gracilis składa się z trzech segmentów: genom jądrowy zawiera 95% DNA, pozostała część znajduje się w chloroplastach i mitochondriach. Ze względu na jego złożony charakter jest on obiektem badań. Wstępne szacunki wskazują na jego bardzo duży rozmiar, nawet do 2 Gbp, występowanie wielu powtarzalnych sekwencji i złożoną i pofragmentowaną strukturę[6].
Charakterystyczne dla Euglenid jest występowanie trzech typów intronów w genach: konwencjonalnych, niekonwencjonalnych i pośrednich. Różnica polega na specyfice granicznych sekwencji zasad azotowych: w wypadku konwencjonalnych jest to GT/C-AG, w wypadku niekonwencjonalnych może być wiele nietypowych sekwencji, pośrednie introny łączą cechy tych grup. W przypadku Eugleny gracilis występują wszystkie trzy typy intronów, czego przykładem jest gen tubA, kodujący tubulinę α. Budowa intronów jest wykorzystywana w ustalaniu filogenezy Euglenid[7].
Orientacja w przestrzeni Eugleny graclilis odbywa się pod wpływem wielu bodźców ze środowiska, np. światło może wywoływać reakcje fotofobiczne, fotokinetyczne, fototaksyczne, dzięki którym organizmy mogą gromadzić się w odpowiednich siedliskach. Reakcje na światło wspomagane są m.in. grawitaksją i aerotaksją. Na podstawie pierwszych obserwacji mikroskopowych rolę fotoreceptora przypisywano stigmie (plamce ocznej), lecz badania wskazały, że właściwym narządem światłoczułym jest ciałko paraksonemalne (PAB, paraflagellar body), zlokalizowane przy krótszej wici, Stigma może pełnić funkcję pomocniczą, jako przesłona ograniczająca ilość światła padającego na fotoreceptor.
Euglena gracilis może używać fizjologicznego receptora grawitacji, dla aktywnej orientacji w przestrzeni w odniesienia do pola grawitacyjnego Ziemi. Sygnał przewodzony jest przy użyciu szlaku składającego się z takich elementów jak mechanoreceptory, jonowe kanały wapniowe, specyficzna kalmodulina (CaM.2), cyklaza adenylowa i kinaza białkowa (PK.4)[8].
E. gracilis jest organizmem modelowym używanym w badaniach naukowych; głównie dzięki swojej dostępności, łatwości hodowli oraz zróżnicowanej biologii. Wyizolowano wiele szczepów tego gatunku, w tym szczepy Z i bacillarus, które stosowane są m.in. w badaniach dotyczących fotosyntezy. Jej popularność jest ograniczona przez brak dokładnych badań genomowych oraz narzędzi do jego manipulacji, ale trwają nad tym prace[6].
